Drug intelligence / Profile preview
[225Ac]Ac-mcp-SC16 is a targeted alpha therapy (TAT) radiopharmaceutical designed for the treatment of neuroendocrine malignancies. It consists of the alpha-emitting radioisotope actinium-225 (225Ac) conjugated to the SC16.56 monoclonal antibody via a macropa-derived (mcp) chelator. The antibody component, SC16.56, specifically targets Delta-like protein 3 (DLL3), a protein frequently overexpressed on the surface of small cell lung cancer (SCLC) and neuroendocrine prostate cancer (NEPC) cells. By delivering high-LET (linear energy transfer) alpha radiation directly to DLL3-expressing cells, the agent induces lethal double-strand DNA breaks while minimizing damage to surrounding healthy tissue. Preclinical data indicates significant antitumor activity, although some renal and ovarian toxicity has been noted in comparative studies.
